Preparation:
Produced from sera of goats immunized with highly pure Recombinant Human IL-17A. Anti-Human IL-17A-specific antibody was purified by affinity chromatography and subsequently biotinylated.

Sandwich ELISA:
For detection of Human IL-17A using sandwich ELISA (100 µL/well antibody solution), use 0.25–1.0 µg/mL of this antibody. When paired with PeproTech Polyclonal Anti-Human IL-17A (500-P07G) as capture antibody, it allows detection of at least 0.2–0.4 ng/well of Recombinant Human IL-17A.

Western Blot:
To detect hIL-17A by Western blot, use 0.1–0.2 µg/mL of this antibody. With compatible secondary reagents, the detection limit for Recombinant hIL-17A is 1.5–3.0 ng/lane under both reducing and non-reducing conditions.

Target Information

Interleukin-17A (IL-17A, also known as CTLA-8) is a cytokine derived from CD4+ T cells that promotes inflammatory responses. It is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection. IL-17A is a 32 kDa disulfide-linked homodimer of 136 amino acids and part of the IL-17 (A–F) family, signaling through the IL-17 receptor (IL-17R/CDw217).

IL-17A enhances ICAM-1 expression in fibroblasts and stimulates epithelial, endothelial, and fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2. It also supports CD34+ progenitor proliferation and neutrophil maturation. IL-17A homologs across species can induce IL-6 secretion in mouse stromal cells, indicating cross-species receptor recognition. IL-17A regulates NF-κB and MAP kinase activity, upregulates IL-6 and COX-2 expression, and enhances nitric oxide production.
