Product Specific Information
The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-linked chemokine receptor binding CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 plays a role in CD4 T cell responses to grafts, with knockout mice showing compromised allograft rejection. Expression occurs on NK cells, subsets of T lymphocytes, Tregs, and Th1-polarized cells. The antibody affects chemotaxis in response to ligand, and ligand presence eliminates staining. In vivo addition delays cardiac and pancreatic islet allograft rejection.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; optimal at ≤0.25 µg per test in 100 µL volume (10⁵–10⁸ cells/test). Titrate for best performance.
Important Notes:
- NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample (regardless of NovaFluor number) before combining with cells.
- For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information
This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC. These chemokines belong to the CXC subfamily, inducing cellular responses like integrin activation, cytoskeletal rearrangement, and chemotaxis. Inhibition by Bordetella pertussis toxin indicates coupling with Gi-subclass G proteins. Expression is high in effector/memory T cells and inflamed tissues, suggesting a role in inflammatory cell recruitment. The receptor is a potential target for small-molecule antagonists in inflammatory disease treatment. Multiple transcript variants exist.
