Product Specific Information

This BB7.2 monoclonal antibody recognizes the human major histocompatibility complex (MHC) class I molecule HLA-A2, a member of the HLA-A family. MHC class I exists as a single alpha chain composed of three domains and associates with beta-2 microglobulin, expressed on all nucleated cells.

The BB7.2 antibody exhibits blocking activity and also recognizes HLA-A28.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.25 µg) per test. A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorophore Information:
PE-eFluor 610 can be excited with laser lines from 488–561 nm and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red equivalent). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

HLA-A is part of the HLA class I heavy chain paralogues. It forms a heterodimer with beta-2 microglobulin, anchored in the membrane. Class I molecules present peptides from the endoplasmic reticulum lumen and are expressed in nearly all cells.

The heavy chain (~45 kDa) gene contains 8 exons:

  • Exon 1: Leader peptide
  • Exons 2–3: α1 and α2 domains (peptide binding)
  • Exon 4: α3 domain
  • Exon 5: Transmembrane region
  • Exons 6–7: Cytoplasmic tail

Polymorphisms in exons 2 and 3 determine peptide binding specificity, commonly typed for bone marrow and kidney transplantation. Hundreds of HLA-A alleles have been described.

HLA and MHC antibodies are critical in Immunopeptidomics, enabling identification and characterization of neoantigens via LC-MS/MS.
