Product Specific Information

This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios.
Helios is a zinc-finger transcription factor similar to Ikaros, expressed in hematopoietic stem cells in both embryo and adult mice.
Highest expression is found in regulatory T cells, while it is not expressed in mature B cells, dendritic cells, or myeloid cells.
Helios forms homodimers and heterodimers with other Ikaros family members to regulate transcription.
Eight isoforms exist, some with dominant-negative functions, upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 also cross-reacts with rat Helios.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using mouse lymph node cells and the Foxp3/Transcription Factor Buffer Set (Product # 00-5523-00).

Recommended usage: 5 µL (0.03 µg) per test (100 µL final volume).
Cell count: 10⁵–10⁸ cells/test.

Fluorochrome Information:
PE-eFluor® 610 excitation at 488–561 nm, emission at 607 nm.
Use a 610/20 band pass filter (PE-Texas Red® equivalent).
Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm / 607 nm / Blue, Green, Yellow-Green Laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Helios is a member of the Ikaros family of zinc-finger proteins (Ikaros, Aiolos, Helios).
These transcription factors regulate lymphocyte development and hematopoietic differentiation.
Helios forms homo- or hetero-dimers with other Ikaros family members.
Multiple transcript variants exist; biological validity of some variants remains unconfirmed.
