Product Specific Information

The ISA-3 monoclonal antibody reacts with human ICOS (Inducible COStimulatory molecule), also known as H4, CRP-1, and AILIM.
ICOS is a T cell-specific activation molecule and a member of the CD28/CTLA-4 family. It plays a key role in T cell activation and humoral immune responses, particularly in memory B cell and plasma cell generation.

• Binding: ICOS binds to ligand B7h/B7RP-1 expressed on activated antigen-presenting cells and inflamed tissues.
• Function: Plate-bound ISA-3 costimulates T cells, inducing IL-4, IL-5, IL-10 production but not IL-2.
• Comparison: Clone C398.4A also cross-reacts with human ICOS but binds a different epitope.
• Recommended for brightest staining: Product #13-9948-82 or Product #12-9948-42.

Applications Reported

• Flow cytometric analysis of stimulated normal human peripheral blood cells.
• Pre-titrated for use at 5 µL (0.25 µg)/test (final volume 100 µL).
• Suitable for 10⁵–10⁸ cells/test.

Fixation

Samples can be stored in:

• IC Fixation Buffer (Product #00-8222), or
• 1-step Fix/Lyse Solution (Product #00-5333)
  for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Filtration

• 0.2 µm post-manufacturing filtered.
