Product Specific Information

The MBSA43 monoclonal antibody reacts with human TIGIT (T cell immunoreceptor with Ig and ITIM domains), also known as VSTM3 or WUCAM. This 26 kDa protein is a member of the poliovirus receptor (PVR) family. TIGIT is expressed on NK cells, regulatory T cells, follicular T helper cells, memory CD4+ and CD8+ T cells, but not on B cells or naive CD4+ T cells. Expression may be induced upon activation.

MBSA43 is reported to cross-react with Rhesus macaque.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume).

Fluorochrome Information:
PE-eFluor™ 610 can be excited with 488–561 nm laser lines and emits at 607 nm. Recommended detection filter: 610/20 band pass (equivalent to PE-Texas Red™). Ensure your instrument supports this fluorochrome.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness or FRET efficiency.

Target Information

TIGIT binds with high affinity to the poliovirus receptor (PVR), leading to increased IL10 secretion and decreased IL12B secretion. It suppresses T cell activation by promoting the generation of mature immunoregulatory dendritic cells.
