Product Specific Information

The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.

  • B cells: Highly expressed in mature plasma cells; regulates differentiation and transcription of B cell-specific genes (e.g., Prdm1/Blimp1).
  • T cells: Implicated in Treg, Th2, Th9, and Th17 cell development and function; upregulated upon activation.
  • Macrophages: Involved in polarization and regulation.
  • Disease relevance: Upregulated in various hematologic malignancies.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Validated using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) according to BestProtocols® “Protocol B: One-step protocol for nuclear intracellular proteins”.
Use ≤0.5 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize concentration empirically.

Target Information

The IRF (interferon regulatory factor) family consists of transcription factors with a unique tryptophan pentad repeat DNA-binding domain. IRFs regulate interferon responses to viral infection and control interferon-inducible genes.
IRF4 is lymphocyte-specific and negatively regulates toll-like receptor (TLR) signaling, central to innate and adaptive immunity. A chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH loci is associated with multiple myeloma. Alternatively spliced transcript variants exist.
