Description:
The eBioMC-631 (MC-631) monoclonal antibody reacts with the carbohydrate moiety galactosylgloboside on Stage-specific embryonic antigen-3 (SSEA-3). Expression is restricted to the surface of human teratocarcinoma stem cells (EC), human embryonic germ cells (EG), and human embryonic stem cells (ES), and is downregulated following differentiation.
Applications Reported:
This antibody has been reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of the 2102Ep cell line.
Recommended ≤ 0.25 µg per test (100 µL final volume).
Cell number should be empirically determined (10⁵–10⁸ cells/test).
Careful titration is recommended for optimal assay performance.
Filtration:
0.2 µm post-manufacturing filtered.
Target Information
SSEA-3 (Stage-specific embryonic antigen 3) is a cell surface antigen expressed along with SSEA-4, TRA-1-60, and TRA-1-81 in embryonic stem cells, embryonal carcinoma cells, and induced pluripotent stem cells (iPS).
In human cells, these markers are downregulated during differentiation, while SSEA-1 is absent in undifferentiated human stem cells and upregulated after retinoic acid-mediated differentiation.
In mouse stem cells, SSEA-1 expression decreases with differentiation, while SSEA-3 and SSEA-4 expression increase.
