Product Specific Information
The TRA-1-60 antibody recognizes a protein expressed on undifferentiated human embryonic stem cells (ES), embryonal carcinoma cells (EC), and embryonic germ cells (EG). The epitope recognized by this antibody is lost upon cell differentiation. Contrary to early reports, TRA-1-60 recognizes a neuraminidase-resistant antigen.
TRA-1-60 specifically binds to a carbohydrate epitope on podocalyxin, a keratan-sulfated glycoprotein and member of the CD34-related family of sialomucins. Podocalyxin is a transmembrane glycoprotein originally identified on glomerular epithelial cells (podocytes) and has been associated with cancer aggressiveness in breast and prostate cancers.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on human embryonal carcinoma (EC) line 2102Ep. Recommended ≤0.25 µg per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Titrate for optimal performance.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
TRA-1-60 is a cell surface antigen expressed along with SSEA-3, SSEA-4, and TRA-1-81 in human embryonic stem cells, embryonal carcinoma cells, and induced pluripotent stem cells (iPSCs). These markers are lost during differentiation, whereas SSEA-1 appears after retinoic acid-mediated differentiation.
