Target Information
KIT (c-KIT) is a proto-oncogene and a type 3 transmembrane receptor for mast cell growth factor (MGF, also known as stem cell factor). KIT was first identified as the cellular homolog of the feline sarcoma viral oncogene v-kit. Together with its ligand, KIT regulates growth and activation of various hematopoietic and non-hematopoietic cells.
Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. Deregulation of KIT receptor tyrosine kinase, particularly by the D816V activation loop mutation, is a key therapeutic target in neoplastic mast cell growth.
c-Kit is expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal tract Cajal cells. Upon binding stem cell factor (SCF), c-kit dimerizes, leading to receptor activation and autophosphorylation of tyrosine residues such as Tyr703. This enables Grb2 docking and activation of the Ras/ERK pathway. SCF/c-kit signaling activates multiple downstream pathways including PI3K, PLC-γ, and JAK/STAT, essential for hematopoiesis, stem cell maintenance, and gametogenesis.
