Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are typically low but play a critical role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, triggering histamine release. IgE is produced by terminally differentiated plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested with normal human peripheral blood cells. Recommended use is 5 µL (0.2 µg) per test in 100 µL final volume. Cell count should be empirically determined (10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

• Updated formulation for optimal NovaFluor staining
• Add 5 µL per sample to antibody cocktails/master mixes

Target Information

IgE is one of five classes of mammalian immunoglobulins, composed of two heavy (epsilon) chains and two light chains. It plays a key role in type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on mast cells and basophils. Crosslinking induces degranulation and release of mediators causing allergic manifestations. IgE also contributes to parasite immunity.
