Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE naturally occurs at low levels in serum and plays a key role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, leading to histamine release and other mediator secretion. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested for flow cytometry using normal human peripheral blood cells. Recommended use is 5 µL (0.2 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL of CellBlox Plus Buffer per sample, regardless of the number of NovaFluor antibodies used.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

IgE is one of five immunoglobulin classes in mammals, composed of two epsilon heavy chains and two light chains. It mediates type I hypersensitivity reactions such as hay fever, asthma, hives, and anaphylaxis by binding to Fc receptors on mast cells and basophils. Crosslinking of IgE triggers degranulation, releasing mediators responsible for allergic symptoms. IgE also contributes to defense against parasitic infections.
