Target Information

Prostaglandins are produced by the metabolism of arachidonic acid. Prostaglandin E2 (PGE2) is one of the five physiologically significant prostanoids. Its diverse physiological and pharmacological effects are mediated through binding to the Prostaglandin E2 receptors (EP1, EP2, EP3, EP4). These effects include modulation of immune, endocrine, cardiovascular, renal, and reproductive systems, as well as smooth muscle regulation.

PGE2 is abundant in the brain, playing a key role in neural functions—especially in newborns—and acts as a mediator of inflammation. PGE2 signals through G-protein coupled EP receptors, which include four subtypes (EP1–EP4). EP2 receptors are 358-amino-acid proteins with a short third intracellular loop, stimulating adenylyl cyclase via Gs coupling and not undergoing PGE2-induced internalization. EP2 receptors contribute to smooth muscle contraction and relaxation, primarily localized in lung, placental, and smooth muscle tissues.
