This antibody is predicted to react with bovine, chicken, canine, amphibian, mouse, and rat based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is breast carcinoma or mantle cell lymphoma.

Target Information

Cyclin D1 (PRAD-1, bcl-1) is a key regulator of the cell cycle, functioning with CDK4 and/or CDK6 to phosphorylate the Rb protein. It acts as a proto-oncogene and is overexpressed in various human neoplasms, including mantle cell lymphomas.
Cyclin D1 regulates protein phosphorylation, mammary epithelial cell proliferation, and adipocyte differentiation.
In humans, the CCND1 gene encoding Cyclin D1 is located on chromosome 11. The protein is expressed in both cytoplasm and nucleus, synthesized during G1 phase, and forms complexes with CDK4 or CDK6 in response to growth factor stimulation.
These complexes inactivate Rb through phosphorylation and modulate CDK inhibitors such as p21Cip1 and p27Kip1.
Without growth factor stimulation, Cyclin D1 is unstable and undergoes ubiquitin-mediated degradation, contributing to G1/S phase arrest.
As a member of the cyclin family, Cyclin D1 shows periodic changes in abundance throughout the cell cycle and plays a central role in regulating CDK activity.
Mutations, amplification, and overexpression of Cyclin D1 are frequently observed in tumors and contribute to tumorigenesis.
Cyclin D1 is a valuable tool for studies in cell cycle biology and cancer research.
