Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is colon carcinoma.

Target Information

ICAD is the inhibitor of caspase-3-activated DNase which controls nuclear apoptosis.
It has two isoforms:

• ICAD-L/DFF45 (45 kDa) – functional isoform
• ICAD-S/DFF35 (35 kDa) – non-functional isoform

Only ICAD-L is functional and inhibits CAD until cleaved by caspase-3 at residues 117 and 224, releasing three fragments.
ICAD also acts as a CAD-specific folding chaperone.
ICAD may be a potential target for restoring normal apoptotic signaling in colon and brain cancer cells.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
