Product Specific Information
This antibody is predicted to react with bovine, mouse, and rabbit based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is colon, lung, or breast carcinoma.
Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a committed step in prostanoid synthesis, including production of inflammatory prostaglandins.
The conversion involves two reactions:
- Cyclooxygenase (COX) reaction converting arachidonate to prostaglandin G2 (PGG2)
- Peroxidase reaction reducing PGG2 to prostaglandin H2 (PGH2)
COX2 is constitutively expressed in some tissues under physiological conditions (endothelium, kidney, brain) and up-regulated under pathological conditions such as cancer and inflammation.
Up-regulation of COX2 is associated with increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 contributes to prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance.
COX2 is naturally inhibited by calcitriol (active Vitamin D) and transcriptionally repressed by glucocorticoids.
It is also a known target of NSAIDs like aspirin, which reduce pain and inflammation driven by COX2.
