Product Specific Information
The sym0F1 monoclonal antibody reacts with human and mouse c-Maf, a 42 kDa basic leucine zipper transcription factor involved in neural, ocular, and hematopoietic systems.
It is crucial for IL-4 expression in Th2 cells and plays roles in Th17 and Tfh cell differentiation by regulating IL-21, IL-23R, and CXCR5 expression.
- Cross-reactivity: Does not cross-react with MafA or MafB.
- Applications Reported: Intracellular staining followed by flow cytometric analysis.
- Applications Tested: Pre-diluted and validated using Th17-polarized mouse splenocytes with Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
- Recommended Use: 5 µL (1.0 µg) per test (final volume 100 µL; 10⁵–10⁸ cells/test).
- Excitation/Emission: 488–561 nm / 607 nm.
- Recommended filter: 610/20 band pass (equivalent to PE-Texas Red).
Light sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57).
Target Information
c-Maf is a basic leucine zipper transcription factor and the cellular counterpart of oncogenic v-maf.
It regulates IL-4 expression in Th2 cells, crystalline gene expression in lens fibers, and insulin gene transcription in pancreatic islets.
c-Maf modulates Th2 differentiation and hematopoietic lineage specification and acts as a transcription factor for IL-10 expression in macrophages.
Chromosomal aberrations involving maf are associated with certain forms of multiple myeloma, and it is expressed in myeloma cell lines and monocytes/macrophages.
