Target Information
TLR5 is a type I membrane protein belonging to the Toll-like receptor family and participates in the innate immune response to microbial agents. It mediates detection of bacterial flagellins and acts via MyD88 and TRAF6, leading to NF-κB activation, cytokine secretion, and inflammatory response.
TLR5 is highly expressed in the liver, detected in the lung, and at very low levels in most other tissues. The TLR5 gene locus is associated with susceptibility to Salmonella. Mouse strains show different resistance to S. typhimurium infection:
- Susceptible: BALB/c, C57BL/6, C3H/He
- Intermediate: DBA/2, C75L
- Resistant: A, CBA
The MOLF/Ei strain is highly susceptible, has a unique TLR5 haplotype, and lower TLR5 expression. A common TLR5 stop codon polymorphism disrupting TLR5 signaling is linked to susceptibility to Legionnaires’ disease, highlighting its importance in innate immunity.
