Product Specific Information

The monoclonal antibody 50-6 recognizes human CD151, also known as PETA-3 (platelet-endothelial cell tetraspan antigen-3).
CD151 (29 kDa) is a member of the tetraspanin family, containing four hydrophobic transmembrane domains, two short cytoplasmic tails, and one small and one large extracellular loop.

CD151 is expressed on:

  • Platelets and megakaryocytes
  • Endothelial cells (at cell-cell junctions)
  • Epithelial cells (basolateral surfaces exposed to extracellular matrix)

Biological role:
Tetraspanins, including CD151, are involved in cell adhesion and migration. CD151 interacts with integrin α3β1 and α6β4, forming complexes with c-Met and integrins in human salivary gland cancer cells. It can also interact with CD46, contributing to early metastasis as a potential positive effector.

The 50-6 monoclonal antibody has been shown to inhibit in vivo metastasis of the human epidermal carcinoma cell line HEp-3.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using human platelets.
Use at 5 µL (0.125 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Target Information

The CD151 gene encodes a member of the transmembrane 4 superfamily (tetraspanins).
These proteins are cell-surface molecules with four hydrophobic domains and are involved in signal transduction regulating cell development, activation, growth, and motility.

CD151 forms complexes with integrins and other tetraspanins, influencing cell adhesion, integrin trafficking, and cell motility.
It enhances invasion and metastasis of cancer cells.
Multiple alternatively spliced transcript variants encoding the same protein have been described.
