Product Specific Information

The monoclonal antibody 1A4 recognizes human, mouse, and rat alpha-smooth muscle actin (α-SMA), a 42 kDa protein that is a major component of the cytoskeletal structural network. This antibody is specific for the alpha form of muscle actin, expressed by smooth muscle cells, myofibroblasts, and myoepithelial cells. It is useful in identifying leiomyomas, leiomyosarcomas, pleomorphic adenomas, and as a prognostic marker for basal cell carcinoma.

Applications Reported:
Used in immunohistochemical staining of frozen and formalin-fixed paraffin-embedded tissue sections, microscopy, and immunocytochemistry.

Applications Tested:

• IHC (P): ≤5 µg/mL (low pH antigen retrieval)
• ICC/IF: ≤5 µg/mL (methanol-fixed and permeabilized cells)
  Careful titration is recommended for optimal assay performance.

Target Information

Smooth Muscle Actin (α-SMA) is part of the actin protein family, essential for cell motility, structure, and integrity. Alpha actin is a major constituent of the contractile apparatus, while beta and gamma isoforms regulate motility. α-SMA is found in smooth muscle vessel walls, gut wall, myometrium, and myoepithelial cells of breast and salivary glands. Defects in the α-SMA gene are associated with familial thoracic aortic aneurysm type 6. During wound healing and fibrocontractive diseases, α-SMA appears in stress fibers of fibroblasts.
