Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells.
SK7 and UCHT1 clones cross-block binding, indicating overlapping epitopes, while OKT3 and SK7 recognize distinct epitopes.
This antibody has been tested for flow cytometric analysis using normal human peripheral blood cells at 5 µL (0.5 µg) per test.

Brilliant Ultra Violet™ 563 (BUV563) emits at 564 nm and is intended for use on cytometers equipped with a 355 nm UV laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.

Light Sensitivity:
This dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation/Emission/Laser:
350 nm excitation; 564 nm emission; Ultraviolet Laser.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR expression.
CD3 gamma, delta, epsilon, and zeta subunits are structurally related members of the immunoglobulin superfamily located on chromosome 11.
CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells.
Defects in the CD3 gene are associated with CD3 immunodeficiency.
