Product Specific Information

Description:
The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes in a developmentally regulated manner and by most peripheral T cells. Crosslinking of the TCR complex with H57-597 can induce T cell activation, proliferation, or apoptosis depending on assay conditions. It is used as a phenotypic marker for TCR beta-expressing T cells.

Flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes at ≤ 0.5 µg/test (100 µL final volume). Optimize antibody titration for best performance.

Fluorophore Information:
Brilliant Ultra Violet™ 496 (BUV496) emits at 496 nm and is suitable for cytometers equipped with a 355 nm UV laser. Confirm instrument compatibility prior to use.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use:

Target Information

T cell receptors (TCRs) enable T cells to distinguish foreign from self-peptides presented by MHC class II molecules, crucial for adaptive immunity. Abnormal TCR recognition of self-peptides is linked to autoimmune diseases and tumor-associated mutant peptides. Superantigen engagement of TCRs can cause toxic shock syndrome. Autoantibodies to TCR V beta segments have been identified in rheumatoid arthritis and systemic lupus erythematosus, potentially modulating inflammatory responses.

Most human T cells express TCR alpha-beta with CD4 or CD8 (single positive), while a small subset lacks both (double negative). Increased alpha-beta DN T cells are found in autoimmune and immunodeficiency disorders. Gamma-delta T cells, primarily epithelial, exhibit limited TCR diversity and distinct antigen recognition, with subsets showing antitumor and immunoregulatory activity.
