Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed on thymocytes and all mature T cells. Binding of 145-2C11 to TCR triggers intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Tested on mouse splenocytes by flow cytometry. Recommended ≤1.0 µg/test in 100 µL final volume. Cell density: 10⁵–10⁸ cells/test. Optimize concentration empirically for best performance.

Fluorophore Information:
Brilliant Ultra Violet™ 563 (BUV563) emits at 564 nm and is designed for cytometers with a 355 nm UV laser. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Cat. SB-4401-42) or Brilliant Stain Buffer™ (Cat. 00-4409-75) to minimize non-specific interactions.

Handling & Fixation:

  • Light sensitive – protect from light.
  • Fixed samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Cat. 00-8222-49) or 1-step Fix/Lyse Solution (Cat. 00-5333-54) with minimal signal loss.

Target Information

The CD3 subunit complex is critical for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, members of the immunoglobulin superfamily encoded on chromosome 11. These subunits have cytoplasmic tails containing tyrosine-based motifs that interact with signal transduction molecules. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Mutations in CD3 genes can result in CD3 immunodeficiency.
