Target Information

The protein tyrosine phosphatase PTPN22 (also known as PTP22, LYP, PEP, formerly PTPN8) is a genetic variant associated with increased risk of autoimmune diseases such as type 1 diabetes and rheumatoid arthritis.
A missense SNP in PTPN22 encodes a hematopoietic-specific phosphatase (Lyp). The risk allele occurs in approximately 17% of Caucasian individuals in the general population and about 28% among those with rheumatoid arthritis.
This variant is believed to disrupt the P1 proline-rich motif important for interaction with the SH3 domain of CSK (cytoplasmic tyrosine kinase), potentially altering the normal regulatory function of these proteins in T cell activation.
The CSK–PTPN22 interaction is highly specific, suggesting that PTPN22 may act as an effector and/or regulator of CSK in T cells and other hematopoietic cells.
