Target Information
LCK는 Src 계열 티로신 키나아제(Src-family tyrosine kinase)에 속하며 T 세포 수용체(TCR) 복합체를 통한 신호 전달에 필수적입니다. TCR이 활성화되면 LCK는 zeta 소단위의 ITAM 모티프를 인산화하여 ZAP70의 SH2 도메인 결합 부위를 형성하고, 이후 ZAP70을 인산화 및 활성화시켜 LAT 어댑터 단백질의 인산화를 통해 후속 신호 전달 플랫폼을 생성합니다.
LCK는 주로 세포막에 위치하지만, 일부는 골지체와 연관되어 약한 TCR 자극 조건에서 Raf 활성화에 기여할 수 있습니다. N-말단에는 myristylation 및 palmitylation 부위가 있으며, PTK 도메인과 SH2, SH3 도메인을 포함하여 인산화 티로신 및 프롤린이 풍부한 모티프와의 단백질-단백질 상호작용을 매개합니다.
LCK는 다양한 자극에 의해 유도되는 세포사멸 조절에도 관여하지만, death receptor에 의한 세포사멸에는 관여하지 않습니다. LCK 관련 질환으로는 Immunodeficiency 22 및 CD45 결핍증이 있으며, 다양한 아이소폼을 암호화하는 대체 스플라이싱 변이체가 보고되어 있습니다.
