Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon stress, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is crucial for p53 activation and stabilization. p53 functions to block proliferation of damaged cells and acts as an anticancer agent. Phosphorylation of Ser392 has been associated with tumor formation. Additionally, p53 is linked to aging, oxidative stress, and learning and memory deficits due to its influence on long-term potentiation (LTP). While p53 levels are low in normal cells, they are elevated in many transformed cell lines, contributing to malignancy. Mutations in p53 disrupt DNA binding and tumor suppressor activity, and germline TP53 mutations are implicated in Li-Fraumeni syndrome.
