Target Information
Huntingtin은 헌팅턴병(Huntington’s disease, HD)과 관련된 유전자 산물로, 선조체 신경세포의 손실을 특징으로 하는 신경퇴행성 질환입니다. 이 질환은 헌팅틴 유전자의 불안정한 삼염기 반복 확장에 의해 발생하며, 단백질 내 폴리글루타민 반복으로 번역됩니다.
HD는 중년기에 발병하는 상염색체 우성 질환으로, 정신 질환, 치매, 불수의적 운동(chorea)이 나타나며 10–20년 내 사망에 이르게 됩니다. 헌팅틴 유전자는 약 180 kb 크기, 67개의 엑손으로 구성되어 있으며, 정상 발달에 필수적입니다.
이 유전자는 두 가지 대체적으로 polyadenylated 형태로 발현되며, 13.7 kb 전사체는 주로 뇌에서, 10.3 kb 전사체는 다양한 태아 및 성체 조직에서 발현됩니다. 헌팅턴병의 유전적 결함은 전사를 완전히 억제하지는 않지만, mRNA의 새로운 특성 부여 또는 단백질 기능 변화를 초래할 수 있습니다.
