Product Specific Information
The IT2.2 monoclonal antibody reacts with human CD86, an ~80 kDa surface receptor also known as B7-2. CD86 and CD80 are members of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells and is upregulated through various stimuli including BCR, CD40, and cytokine receptors.
CD86 interacts with CD28 and CTLA-4 on T cells, playing a key role in T-B cell crosstalk, T cell costimulation, and antibody production. Its upregulation kinetics support its major role in the primary immune response phase.

- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry on human peripheral blood cells. Use 5 µL (0.05 µg)/test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.
Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
- Required for optimal staining with NovaFluor conjugates
- Add 5 µL per sample (regardless of number of NovaFluors)
- For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells)
NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information
CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation during antigen presentation. Inadequate co-stimulation via CD86/CD80 can induce tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed by antigen-presenting cells.
Binding with CD28 provides activation signals, while binding with CTLA-4 inhibits T-cell activation. Alternative splicing produces multiple isoforms of CD86.
