Product Specific Information

The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes and a subset of mature T cells (T-helper cells), and at low levels on monocytes. CD4 serves as a receptor for HIV, and RPA-T4 blocks HIV binding and mixed lymphocyte reaction. It recognizes a different epitope than the OKT4 antibody.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology, engineered nucleic acid dye structures offering optimized fluorescent signatures with minimal spillover and spread.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. Expressed mainly in T-helper lymphocytes, as well as monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of the brain and gut. CD4 facilitates early T-cell activation via association with the TCR complex and tyrosine kinase Lck and may mediate neuronal damage in immune-related CNS diseases. Multiple spliced transcripts have been identified (RefSeq, July 2017).
