Product Specific Information

The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor belonging to the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated through various stimuli such as BCR complex, CD40, and cytokine receptors.
CD86 interacts with CD28 and CTLA-4 on T cells, playing a critical role in T-B cell crosstalk, costimulation, and immune regulation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.2 µg) per test in a final volume of 100 µL.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer Usage:
Add 5 µL per sample to antibody cocktails prior to combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology based on engineered nucleic acid dye structures that minimize spillover and spread.

Target Information

CD86 is a costimulatory molecule that interacts with CTLA-4 and CD28 on T cells. It plays a vital role in antigen presentation and T-cell activation. Insufficient co-stimulation via CD86/CD80 can induce tolerance. CD86 is expressed on antigen-presenting cells and is involved in immune regulation.
Diseases associated with CD86 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
