Product Specific Information

The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor. CD86 and CD80 are members of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated by stimuli such as BCR complex, CD40, and cytokine receptors.
CD86 interacts with T cell surface molecules CD28 and CTLA-4, playing a critical role in T-B crosstalk, T cell costimulation, and antibody production.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry on human peripheral blood cells (5 µL per test, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample.

Target Information

CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule during antigen presentation. Insufficient co-stimulation can induce tolerance. CD86 is a type I membrane protein in the immunoglobulin superfamily, expressed by antigen-presenting cells.
Binding with CD28 activates T cells, while binding with CTLA4 suppresses activation and immune response.
Alternative splicing produces multiple isoforms. CD86 dysfunction is associated with gallbladder squamous cell carcinoma and myocarditis.
