Product Specific Information

The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor belonging to the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated upon stimulation via BCR complex, CD40, or cytokine receptors. It interacts with CD28 and CTLA-4 on T cells, playing a key role in T-B cell crosstalk, costimulation, and immune response regulation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells at 5 µL/test (0.2 µg).
Each test stains a sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells.
• NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures with optimized spillover and spread control.
• Excitation: 552 nm; Emission: 690 nm; Recommended Laser: 561 nm (Yellow).

Target Information

CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation during antigen presentation. Insufficient co-stimulation can lead to immune tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed on antigen-presenting cells. Binding with CD28 activates T cells, while binding with CTLA-4 downregulates activation. Alternative splicing produces multiple isoforms. Dysfunction of CD86 is associated with gallbladder squamous cell carcinoma and myocarditis.
