Product Specific Information

Description:
The 6H6 monoclonal antibody reacts with human CD123, the alpha chain of the IL-3 receptor. This 60–70 kDa transmembrane protein binds IL-3 with low affinity by itself and with high affinity when associated with CD131 (common beta chain). CD123 is expressed by myeloid precursors, macrophages, dendritic cells, mast cells, basophils, and megakaryocytes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
The 6H6 antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤1 µg per test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

NovaFluor dye compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample (regardless of number of NovaFluors). For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates are based on Phiton™ technology using novel nucleic acid dye structures engineered for optimized fluorescent signatures with reduced spillover and spread.

Target Information

IL-3Ra (Interleukin-3 receptor subunit alpha):
Belongs to the type I cytokine receptor family and forms a heterodimer with the common beta (CDw131) subunit. Responsible for IL-3 signal transduction. Overexpressed on acute myeloid leukemia (AML) blasts compared with normal hematopoietic cells, making it a potential therapeutic target.
