Product Specific Information
This antibody is predicted to react with porcine based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is placenta or breast carcinoma.
Target Information
MMP9 (matrix metallopeptidase 9, GELB, CLG4B) is a matrix metalloproteinase belonging to a family of zinc and calcium-dependent endopeptidases that degrade extracellular matrix proteins.
MMP9 is secreted as a 92 kDa zymogen; cleavage of pro-MMP9 yields the active enzyme (82 kDa).
It contains:
- Gelatin-binding domain (three fibronectin type II units)
- Catalytic domain with zinc-binding site
- Proline-rich type V collagen-homologous domain
- Hemopexin-like domain
MMP9 is produced by monocytes, macrophages, neutrophils, keratinocytes, fibroblasts, osteoclasts, and endothelial cells.
It is involved in inflammatory responses, tissue remodeling, wound healing, tumor growth, and metastasis.
MMP9 contributes to skin carcinogenesis and degrades type IV and V collagens.
Studies indicate roles in IL-8-induced mobilization of hematopoietic progenitor cells, tumor-associated tissue remodeling, idiopathic atrial fibrillation progression, and aortic aneurysm formation.
