Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature. A suggested positive control is placenta tissue.
Target Information
Matrix metalloproteinases (MMPs) are enzymes involved in extracellular matrix degradation. Sixteen members are known, with ten existing in soluble form and MT-MMPs as integral membrane proteins. MT1-MMP, MT2-MMP, and MT3-MMP (also known as MMP14, MMP15, and MMP16, respectively) contain a C-terminal transmembrane domain anchoring them to the cell surface and an 8-amino-acid insert in their catalytic domain. MT1-MMP activates progelatinase A (MMP-2) and progelatinase B. MT2-MMP plays a minor role in activating pro-MMP-2 in breast carcinomas. Coexpression of MT1-MMP and MT2-MMP in advanced breast carcinomas suggests involvement of MT2-MMP in metastasis.
