Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16A (FcγRIIIA) is expressed on macrophages and NK cells, while CD16B (FcγRIIIB) is expressed on neutrophils.
Two polymorphisms (NA-1, NA-2) exist for CD16B; NA-2 homozygotes show lower phagocytic capacity.

CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, and mediates ADCC, phagocytosis, degranulation, and proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for use at 5 µL (0.6 µg) per test (100 µL total volume).
Recommended cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD16 encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance, ADCC, and viral infection enhancement.
The gene FCGR3A (chromosome 1) is expressed on NK cells (transmembrane form), while FCGR3B is expressed on neutrophils (PI-anchored form).
Mutations are associated with Immunodeficiency 20, Herpes Zoster, and susceptibility to SLE and neonatal neutropenia.
Alternative splicing produces multiple isoforms.
