Product Specific Information

The eBioCB16 monoclonal antibody recognizes human CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 exists as two forms:

• CD16A (FcγRIIIA): Found on macrophages and NK cells (hetero-oligomeric, polypeptide-anchored)
• CD16B (FcγRIIIB): Found on neutrophils (monomeric, GPI-anchored)

Two polymorphisms, NA-1 and NA-2, affect phagocytic capacity. CD16 binds IgG immune complexes (preferentially IgG1, IgG3) and initiates signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells.
Use 5 µL (0.4 µg) per 100 µL of cell suspension (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• Always include CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) in antibody cocktails for optimal staining.
  • Add 5 µL per sample (regardless of number of NovaFluor dyes).
  • For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures for optimized fluorescence with reduced spillover and spread.
