Product Specific Information

This HP-3G10 monoclonal antibody reacts with CD161 (also known as NKR-P1A), a member of the C-type lectin superfamily. CD161 is expressed on natural killer cells and various T cell subsets, including memory/effector CD4+ and CD8+ T cells, γδ TCR T cells, and a subset of CD3+ thymocytes. CD161 expression has also been demonstrated on human Th17 CD4+ T cells. Lectin-like transcript-1 (LLT1), also known as osteoclast inhibitory lectin, has been identified as the ligand for CD161.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

eFluor™ 450 is an alternative to Pacific Blue™ and emits at 445 nm when excited with the Violet laser (405 nm).

Target Information

KLRB1 (Killer cell lectin-like receptor subfamily B, member 1, CD161) is a type II membrane protein expressed by NK cells and a subset of CD3⁺ T cells. It forms a homodimeric structure (40–44 kDa) and plays an inhibitory role on NK cells. Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide, activating AKT1/PKB and RPS6KA1/RSK1 kinases, leading to T cell proliferation. KLRB1 binds CLEC2D/LLT1 and inhibits NK cell-mediated cytotoxicity and interferon-gamma secretion.
