Product Specific Information

The monoclonal antibody HP-MA4 recognizes several CD158 proteins (KIR2D family), specifically KIR2DL1 (CD158a), KIR2DS1 (CD158h), KIR2DS3, and KIR2DS5 (CD158g).
It does not recognize KIR2DL2 (CD158b1), KIR2DS2 (CD158j), KIR2DL4 (CD158d), or KIR2DS4 (CD158i).
Cross-reactivity to KIR2DL3 (CD158b2) and KIR2DL5 (CD158f) is unknown.

CD158 proteins are members of the killer cell immunoglobulin-like receptor (KIR) family, expressed on NK cells and subsets of T cells.
Each KIR has either a short (S) or long (L) cytoplasmic domain, mediating activating or inhibitory signals, respectively.
Cross-reactivity of HP-MA4 to KIR2DS3 and KIR2DS5 was published in 2013.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells.
Recommended use: 5 µL (0.125 µg) per test in a final volume of 100 µL.
Cell number can range from 10⁵ to 10⁸ cells/test.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emits at 445 nm.
Ensure the instrument supports detection of this fluorochrome.

Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Killer cell immunoglobulin-like receptors (KIRs) are transmembrane glycoproteins expressed by natural killer (NK) cells and subsets of T cells.
KIR genes are polymorphic and clustered on chromosome 19q13.4 within the leukocyte receptor complex (LRC).
KIR proteins are classified by:

  • Number of extracellular immunoglobulin domains (2D or 3D)
  • Cytoplasmic domain length (L or S)

KIRs with long cytoplasmic domains transduce inhibitory signals via ITIM motifs, while those with short domains associate with TYRO protein tyrosine kinase binding protein to transduce activating signals.
Their ligands are subsets of HLA class I molecules, playing key roles in immune regulation.
