The VK9 monoclonal antibody recognizes human Globo H, a hexasaccharide derived from the pentasaccharide precursor SSEA3 (stage-specific embryonic antigen 3). Globo H belongs to a family of antigenic carbohydrates expressed on cancer cell surfaces as glycolipids and possibly glycoproteins.

Globo H is overexpressed in multiple carcinomas including breast, colon, ovarian, gastric, pancreatic, lung, and prostate cancers. Expression is found in more than half of ductal, lobular, and tubular breast carcinomas, but not in non-epithelial breast tumors. It is also expressed on cancer stem cells and human embryonic stem cells, with expression decreasing upon differentiation toward neural progenitor or endodermal lineage cells. In normal tissues, expression is limited to cells composing secretory borders.

Globo H is considered a potential immunotherapy target for various epithelial cancers.

Applications Reported:

• Immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections
• Microscopy

Applications Tested:

• Tested by IHC of formalin-fixed paraffin-embedded human tissue using high or low pH antigen retrieval at ≤5 µg/mL.
• Tested by Western blot of reduced MCF-7 cell lysate at ≤5 µg/mL.
• Recommended to titrate for optimal assay performance.

Target Information

Globo H is a hexasaccharide formed from the pentasaccharide precursor SSEA3. It is expressed on the surface of cancer cells and is overrepresented in many carcinomas. Expression is limited in normal tissues and is considered an important target for cancer immunotherapy.
