The monoclonal antibody 7UNA8U6 reacts with human, mouse, and rat TdT (terminal-deoxynucleotidyl transferase), a 60 kDa polymerase responsible for template-independent addition of N-nucleotides at gene segment junctions of developing lymphocytes.
Expression is found in the nucleus of immature lymphocytes but not in mature lymphocytes or non-lymphoid cells. TdT plays a critical role in TCR and Ig gene rearrangement, increasing lymphocyte repertoire diversity through addition of N-nucleotides.
After positive selection, TdT expression decreases or becomes absent. TdT expression has been observed in lymphoma or lymphoblastic leukemia cells.

• Immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections

Applications Tested:

• Immunohistochemistry (human and mouse tissue, low pH antigen retrieval): ≤10 µg/mL
• Western blot (mouse and rat cell lysate): ≤5 µg/mL
  It is recommended to titrate the antibody for optimal performance.

Target Information

Terminal Deoxynucleotidyl Transferase (TdT) is a DNA polymerase located in the cell nucleus that catalyzes polymerization of deoxynucleotides at 3′ hydroxyl ends of oligo- or polydeoxynucleotide initiators, functioning without a template.
TdT is a highly specific marker for diagnosis and classification of acute lymphoblastic lymphoma/leukemia. Determination of TdT expression is valuable for differentiating lymphoblastic lymphoma from Burkitt’s lymphoma.
