Product Specific Information

The NP-6G4 monoclonal antibody reacts with human CD195 (CCR5), an approximately 45 kDa member of the G protein-coupled receptor (GPCR) superfamily that binds chemokines including MIP-1α, MIP-1β and RANTES.
Human CCR5 is expressed on resting T cells with a memory/effector phenotype, monocytes, macrophages, and immature dendritic cells. CCR5 functions as a receptor to direct lymphocyte chemotaxis and serves as the primary co-receptor for macrophage-tropic HIV-1, binding through gp120.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Recommended staining: 37°C for 1 hour in Flow Cytometry Staining Buffer.
Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.

Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
Excitation: 488–561 nm
Emission: 775 nm

Target Information

HIV and related viruses require coreceptors in addition to CD4 for infection. Among the chemokine receptor family, CCR5 acts as a principal coreceptor for macrophage- and dual-tropic HIV-1 strains, facilitating fusion and entry into human white blood cells.
CCR5 is required for infection by HIV-1, HIV-2, and SIV. Beta-chemokines RANTES, MIP-α, and MIP-β bind CCR5 and inhibit M-tropic HIV-1 infection. CCR5 associates with the CD4-gp120 complex, enabling membrane fusion and virus entry. CCR5 mRNA is expressed in lymphoid organs and monocytes.
