Product Specific Information

This P84 monoclonal antibody reacts with mouse CD172a (SIRPα), a cell surface glycoprotein with three Ig-like extracellular domains and two cytoplasmic ITIM motifs that recruit SHP-1 and SHP-2. CD172a is expressed on monocytes, macrophages, and dendritic cells, but not on T or B lymphocytes. It is also found in neurons and other CNS tissues. The extracellular ligand for CD172a is CD47.

CD172a is involved in dendritic cell-mediated T cell activation, neutrophil migration, and phagocytosis. This antibody has reported neutralizing activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse bone marrow cells at ≤1.0 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize concentration empirically.

Light Sensitivity: Tandem dye sensitive to photo-induced oxidation. Protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information

SIRPα (CD172a, signal-regulatory protein alpha) is a receptor-type transmembrane glycoprotein expressed on myeloid cells such as granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells. It acts as a substrate for activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase signaling.

Binding of SIRPα to CD47 results in phosphorylation of ITIM motifs, recruiting SHP-1 and SHP-2 phosphatases. This interaction regulates phagocytosis inhibition, T cell and DC activation, and other physiological processes. Cancer cells often overexpress CD47, activating SIRPα to evade macrophage-mediated destruction.
