Product Specific Information

The APA5 monoclonal antibody reacts with the mouse CD140a molecule, the alpha chain of the platelet-derived growth factor receptor (PDGF receptor). PDGFRα is a receptor tyrosine kinase that forms dimers upon ligand binding, activating phosphorylation of substrates. Dimers can be alpha/alpha, beta/beta, or alpha/beta combinations, serving as substrates for kinase activity. CD140a is expressed in embryonic tissues and mesenchymal-derived adult mouse cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of NIH/3T3 cells. Recommended ≤1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize by titration for best performance.

Fluorochrome Information:
Super Bright 436 is excited with a 405 nm violet laser and emits at 436 nm. Recommended detection using a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use the Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

PDGFRA (platelet-derived growth factor receptor alpha) is a cell surface receptor tyrosine kinase that binds PDGF family members, which are mitogens for mesenchymal-origin cells. The receptor forms homo- or heterodimers (alpha/alpha or alpha/beta), depending on the bound ligand. Knockout mouse studies show PDGFRA is essential for kidney development, as heterozygous mice exhibit kidney defects.
