Product Specific Information
The APB5 monoclonal antibody reacts with the mouse CD140b molecule, the beta chain of the platelet-derived growth factor receptor (PDGF receptor). PDGFRb is a receptor tyrosine kinase that forms dimers upon ligand binding and phosphorylates substrates. Dimers may be alpha/alpha, beta/beta, or alpha/beta combinations, serving as substrates for kinase activity.
CD140b is expressed in embryonic tissues and mesenchymal-derived cells of adult mouse tissues. PDGFR beta plays a significant role in fibrous atherosclerotic lesion formation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on NIH/3T3 cells at ≤1.0 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Titrate for optimal performance.
Super Bright 436 is excited at 405 nm (violet laser) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
PDGFRB is a receptor tyrosine kinase implicated in atherogenesis and cell transformation by the SIS oncogene. These growth factors act as mitogens for mesenchymal-origin cells. The growth factor bound determines whether the receptor forms alpha/alpha, beta/beta, or alpha/beta dimers. The PDGFRB gene is located on chromosome 5 near granulocyte-macrophage colony-stimulating factor and macrophage-colony stimulating factor receptor genes, all associated with 5-q syndrome. A translocation between chromosomes 5 and 12 involving PDGFRB and ETV6 leads to chronic myeloproliferative disorder with eosinophilia.
