Product Specific Information
This 4B4 (4B4-1) monoclonal antibody reacts with human CD137 (4-1BB, TNFRSF9), an inducible member of the TNFR family expressed on T cells, NK cells, dendritic cells, granulocytes, and mast cells.
CD137 signaling recruits TRAF1 and TRAF2 and is involved in T cell activation, survival of activated and CD8 memory T cells, and suppression of myelopoiesis and dendritic cell development.
Stimulation promotes expansion of CD4⁺CD25⁺ T regulatory cells ex vivo.
The ligand 4-1BBL is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry of stimulated human peripheral blood cells.
Recommended use: 5 µL (0.5 µg) per test in a final volume of 100 µL.
Cell number: 10⁵–10⁸ cells/test (empirical determination required).
Fluorochrome Information:
Super Bright 436 can be excited with a 405 nm violet laser and emits at 436 nm. Recommended detection filter: 450/50 bandpass or equivalent.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
CD137 (4-1BB, TNFRSF9) is an inducible costimulatory molecule mainly expressed on activated T cells.
Its ligand, 4-1BBL, is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.
CD137 signaling supports activated and memory CD8⁺ T cell survival, T cell expansion, and regulation of cytokine release affecting monocyte survival.
Soluble CD137 may act as a negative regulator in immune responses.
