Product Specific Information

By Western blot, PA1-16817 detects heterogeneously glycosylated NPC1 protein with prominent bands at approximately 170 and 220 kDa.
Suggested positive control: Human fibroblast cell lysate.

Target Information

Mutations in the Niemann-Pick disease type C1 (NPC1) gene cause a fatal progressive neurodegenerative disorder characterized by abnormal lipid sequestration, including cholesterol and glycosphingolipids.
The NPC1 protein is a large membrane protein located in endosomes and lysosomes, mediating intracellular cholesterol trafficking by binding cholesterol at its N-terminal domain.
NPC1 transports low-density lipoproteins to late endosomal/lysosomal compartments for hydrolysis and cholesterol release.
Recent studies have identified NPC1, along with FTO, MC4R, and PTER, as a risk locus for early-onset and morbid adult obesity in European populations.
