Product Specific Information

The monoclonal antibody mhalx21 reacts with mouse interleukin-21 (IL-21), a 17 kDa immunomodulatory cytokine produced mainly by NKT, Th17, and T follicular helper (Tfh) cells.
In Tfh cells, IL-21 expression leads to autocrine signaling through IL-21R and STAT3, activating Bcl6 and promoting effector function. IL-21 is essential for T cell-dependent B cell differentiation, IgG production, and induction of apoptosis in naive B cells.

In Th17 cells, IL-21 expression and feedback through STAT3, IRF4, and RORγt upregulate IL-23R, supporting Th17 maturation and maintenance. IL-21 also downregulates Foxp3. Elevated IL-21 levels are found in chemically-induced colitis, while IL-21-deficient mice show protection due to reduced Th17-associated gene expression.

Preliminary data indicate that clone mhalx21 recognizes a different epitope than anti-mouse IL-21 clone FFA21 (Product #51-7211).

Tested by intracellular staining of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended use: ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Antibody titration is recommended for optimal performance.

Target Information

IL-21 is a 17 kDa cytokine produced by NKT, Th17, and Tfh cells. It mediates autocrine signaling via IL-21R and STAT3, promoting transcriptional activation by Bcl6 and supporting Tfh cell development and effector function.
IL-21 regulates B cell differentiation, IgG production, and apoptosis in naive B cells.
In Th17 cells, IL-21 enhances IL-23R expression and downregulates Foxp3.
IL-21-deficient mice are resistant to chemically induced colitis, while elevated IL-21 levels are observed in colitis models.
