SMAD3 belongs to the SMAD protein family, homologous to the Drosophila mothers against decapentaplegic (Mad) and C. elegans Sma genes.

SMAD3 is activated by transforming growth factor-beta (TGF-β) and plays a role in transcriptional regulation and carcinogenesis.
