Target Information
STAT3 (Signal Transducer and Activator of Transcription 3) is a transcription factor activated by cytokines and growth factors. It is phosphorylated by receptor-associated kinases, translocates to the nucleus, and regulates gene expression.
- Activation by: IFN-α, IFN-γ, EGF, PDGF, IL-6
- Phosphorylation at Tyr705 by JAK1/JAK2 is essential for dimerization and DNA binding
- Located on chromosome 17q in humans
- Plays a key role in cell growth, apoptosis, and transcriptional regulation
- Interacts with coactivators such as CREB-binding protein and transcription factors c-Jun and CREB
- PIAS3 acts as a specific inhibitor of STAT3
- Three alternatively spliced transcript variants exist
- Knockout of STAT3 in mice is embryonically lethal
